Vital Staining of Damaged Liver Cells

چکیده

Some dyes are excreted almost exclusively by the liver and the rate of their elimination from the circulating blood is used as a test of certain aspects of hepatic function in man. Rose bengal is one of these dyes,""a"' and the test in which it is used is similar to the better known procedure utilizing bromsulphalein. Clearance of rose bengal from the blood is known to be retarded in hepatic diseases of man9 and in chloroform-induced liver damage in dogs.' But morphological aspects of hepatic excretion of rose bengal and other "liver function" dyes have received only slight attention10'1' (although the hepatic excretion of fluorescein, a related dyestuff, has been studied by means of ultraviolet microscopy7). This report presents an account of the intracellular deposition and clearance of rose bengal in normal and carbon tetrachloride-damaged livers of mice.

Background and Purpose: Perfused rat liver and isolated hepatocytes are useful to study specific metabolism of this organ. Traditionally, mechanical, chemical and enzymatic methods have been used to isolate hepatocytes, and it appears that a mixed method is the preferred choice. Materials and Methods: The rat liver perfused in situe in three subsequent steps, 10 minutes with Krebs-Ringer s...

In vitro differentiation of rat mesenchymal stem cells to hepatocyte lineage

Objective(s): Mesenchyme is a type of undifferentiated loose connective tissue that is derived mostly from mesoderm. Recently, mesenchymal stem cells (MSCs), as adult stem cells (ASCs) able to divide into a variety of different cells, are of utmost importance for stem cell research. In this research, ability of the liver extract to induce differentiation of rat derived omentum tissue mesenchyma...

Genetic Abolishment of Hepatocyte Proliferation Activates Hepatic Stem Cells

Quiescent hepatic stem cells (HSCs) can be activated when hepatocyte proliferation is compromised. Chemical injury rodent models have been widely used to study the localization, biomarkers, and signaling pathways in HSCs, but these models usually exhibit severe promiscuous toxicity and fail to distinguish damaged and non-damaged cells. Our goal is to establish new animal models to overcome thes...

Recent Technological Advances in Hepatogenic Differentiation of Stem Cells Relevant to Treatment of Liver Diseases

Liver failure, in an acute or chronic form, is a growing health problem ranking as one of the leading causes of death worldwide. Inborn errors of metabolism characterized by defects in hepatic enzymes or other proteins with metabolic functions, such as receptors or transporters accompanied with environmental factors involve etiology and presentation of liver failure. Currently, the only establi...

Isolation and Phenotyping of Normal Mouse Liver Dendritic Cells by an Improved Method

Introduction Dendritic cells (DCs) are bone marrow-derived cells, which migrate to lymphoid and non-lymphoid organs via blood. Liver DCs are believed to play an important role in the regulation of hepatic allograft acceptance. However, because of inherent difficulties in isolating adequate numbers of DCs from liver, limited information is available on the phenotype and functions of liver DCs. ...
